I was just leaving Austria, heading to Gyor, about at this coordinates, two trucks left the highway for entering the gas station. Both trucks dissapeared when they've left the highway. I am not sure if this happens to cars too, but I guess it does.
I have no traffic mods in use, never used any on this savegame.

L.E: I've double checked, been watching the traffic flow. Only the trucks are getting despawned, check this short gallery. Cars and buses don't get despawned. I can't confirm what happens to trucks w/o trailer, as only trucks w/ trailer passed around while I was watching.
Keep your eyes on the blue car behind the red DAF. The car has traveled like 5 extra meters between the 2 pictures. So the DAF shd've been still in sight, but it is not, It has just despawned as soon as he passed through the traffic signs.
